该剧讲述的是美丽的苗族姑娘阿榜回到了“歌与舞的海洋、情与爱的世界”的故乡，与妹妹阿莆在“姊妹节”送出了“姊妹饭”，姐妹俩心爱的人识破了附加在“姊妹饭”里的“一对筷子”的无言情书。从小在沿海城市深圳长大的阿榜不仅回到了苗族山乡，而且回归了苗族文化。

描写的是一对孪生姊妹（胡蝶兼饰）的不同遭遇。妹妹二宝自幼跟随私贩洋枪的父亲逃往城市，长大以后被父亲送给了军阀钱督办做姨太太；姐姐大宝则和母亲（宣景琳饰）一起住在乡下，后来嫁给一个穷苦的木匠桃哥（郑小秋饰），父母姊妹不通音讯，彼此不知下落。十多年后，大宝夫妇和母亲从乡下流落到城市。桃哥去做工，大宝抛下未满月的孩子，到钱督办的公馆去当奶妈，她带的正好是二宝的孩子。二宝作为主妇，对大宝非常冷淡，甚至因为大宝外貌酷似自己而感到反感；大宝为了保住饭碗，只好尽心服侍小主人。一天，桃哥干活时摔伤，无钱医治，大宝便恳求二宝预支一点工钱，但二宝非但不肯，反而打了大宝一记耳光。大宝为了救治丈夫，便窃取了小主人的金锁片，不巧又被钱督办的妹妹撞见，大宝失手碰倒花瓶，致使钱妹遭误伤而亡，大宝即以杀人罪入狱。大宝入狱后，母亲前来探监，才发现主办大宝一案的军法处长正是遗弃自己多年的丈夫。丈夫在妻子的愤怒斥责和要求下，安排妻子和大宝、二宝会面。真相大白后，二宝终于为母亲和姐姐的苦难遭遇所感动，不顾父亲劝阻，带领她们驱车前往钱督办处求情。1934年，《姊妹花》被评选为最佳有声片；1935年，《姊妹花》曾和《渔光曲》等影片一起被推荐参加莫斯科电影节展映。
